The Polycomb repressive complex 2 (PRC2) is a conserved chromatin-remodelling that catalyses the trimethylation of histone H3 lysine 27 (H3K27me3), mark associated with gene silencing. PRC2 regulates chromatin structure and expression during organismal tissue development homeostasis in adult. core subunits are various accessory proteins modulate its function recruitment to target genes. multimeric composition results two distinct variant complexes PRC2, PRC2.1 PRC2.2. Metal response element-binding transcription factor (MTF2) one Polycomb-like (PCLs) forms complex. MTF2 highly conserved, as an subunit it has important roles embryonic stem cell self-renewal differentiation, development, cancer progression. Here, we review impact assembly, catalytic activity, spatiotemporal function. emerging paradoxical evidence suggesting divergent either tumour suppressor or oncogene different tissues merits further investigations. Altogether, our illuminates context-dependent group (PcG) protein-mediated epigenetic regulation. Its on disease paves way for deeper understanding regulation novel therapeutic strategies.
